炎症性肠病(IBDs)是一种全球性疾病,发病率在发展中国家呈上升趋势,该病常反复发作,严重影响患者的正常生活。生物钟系统生物体内生的振荡器,控制着多种生理行为进程,生物钟与炎症性疾病关系密切,生物钟还可能是炎症性疾病症状产生昼夜差异的重要因素。尽管昼夜节律如何影响炎症越明了,但炎症调节昼夜节律的分子机制仍然难以捉摸。LncRNA(长非编码RNA)是一类非编码RNA,可通过核苷酸碱基配对或RNA折叠产生的结构域的形成,与DNA,其他RNA和蛋白质发生相互作用,从而调节基因转录。一些lncRNA与免疫反应有关,在炎症反应(如内毒素血症和肾炎)的发生中发挥作用。另外,lncRNA对生物钟有潜在的影响,还可能受生物钟的控制。然而,lncRNA在结肠炎与昼夜节律的交互作用中的作用在很大程度上是未知的。


近期暨南大学吴宝剑教授团队在Science Advances上发表了题为” An NF-κB-driven lncRNA orchestrates colitis and circadian clock ”的研究论文,该研究发现NF-κB驱动的一种周期性表达的lncRNA——Lnc-UC,能通过表观遗传机制调控有抗炎作用的节律基因Rev-erbα的表达,从而将昼夜节律和结肠炎联系起来。靶向Lnc-UC可能是干预炎症与生物钟之间互相作用控制结肠炎的新策略
